Spain keeper David De Gea could be a doubt for their other World Cup Group B games after suffering a muscle injury.
The Manchester United player, 23, missed training on Sunday after picking up a buttocks injury.
The champions face Chile on Wednesday and then Australia five days later.
A Spain statement read: "The Spanish team medical staff will assess the Madrid-born keeper's progress on a daily basis to determine when he can rejoin the group."
Iker Casillas is Spain's first-choice keeper and played during their 5-1 defeat by Netherlands on Friday.
Spain's other goalkeeper is Pepe Reina, who spent last season on loan at Napoli from Liverpool.
